چکیده

Hypochlorous acid (HClO) is a common reactive oxygen species (ROS), with high chemical reactivity. Myeloperoxidase (MPO) an enzyme that catalyzes in vivo redox reactions between H2O2 and Cl− to produce HClO. Abnormal levels of HClO MPO may lead oxidative stress, irreversible tissue damage and, thus, serious diseases; they are thus becoming important biomarkers therapeutic targets. In this work, using HClO-induced site-specific cleavage phosphorothioate-modified DNA trigger rolling circle amplification (RCA), RCA-assisted biosensors have been developed for the highly sensitive specific detection MPO. Only two oligonucleotides used sensing systems. The powerful signal-amplification capability RCA endows systems sensitivity, fluorescent response thioflavin T (ThT) G-quadruplexes products makes label-free signal output possible. proposed were demonstrated work well not only quantitation limits 1.67 nM 0.33 ng/mL, respectively, but also screening inhibitory capacity evaluation inhibitors, holding great promise disease diagnosis drug analysis.

Rolling Circle Amplification (RCA): an approach for quick detection and identification of fungal species

Conventional methods for fungal identification in the clinical laboratory rely on morphological andphysiological tests. These tests often need several days or weeks to complete and are frequentlyunspecific. Molecular identification mostly implies sequencing, which is relatively expensive andtime-consuming, as well as impractical for large numbers of isolates. The Rolling CircleAmplification app...

Gold Nanowire Based Electrical DNA Detection Using Rolling Circle Amplification

We present an electrical sensor that uses rolling circle amplification (RCA) of DNA to stretch across the gap between two electrodes, interact with metal nanoparticle seeds to generate an electrically conductive nanowire, and produce electrical signals upon detection of specific target DNA sequences. RCA is a highly specific molecular detection mechanism based on DNA probe circularization. With...

Sensitive isothermal detection of nucleic-acid sequence by primer generation–rolling circle amplification

A simple isothermal nucleic-acid amplification reaction, primer generation-rolling circle amplification (PG-RCA), was developed to detect specific nucleic-acid sequences of sample DNA. This amplification method is achievable at a constant temperature (e.g. 60 degrees C) simply by mixing circular single-stranded DNA probe, DNA polymerase and nicking enzyme. Unlike conventional nucleic-acid ampli...

Ultrasensitive colorimetric DNA detection using a combination of rolling circle amplification and nicking endonuclease-assisted nanoparticle amplification (NEANA).

A combination of rolling circle amplification and nicking endonuclease-assisted nanoparticle amplification (NEANA) is used for the rapid, colorimetric detection of DNA. The integration of rolling circle amplification into the NEANA approach allows for detection of oligonucleotides with arbitrary sequences at ultralow concentrations.

Circle-to-circle amplification for precise and sensitive DNA analysis.

We present a tightly controlled process for strand-specific amplification of circularized DNA molecules. Tandem repeated complements of DNA circles are generated by rolling-circle replication, and converted to monomer circles of opposite polarity to that of the starting material. These circles are then subjected to one more round of rolling-circle replication and circularization, and the proces...
